A Cryostat is a device used to keep samples or mounted equipment at extremely low cryogenic temperatures.  It can be kept at extremely low temperatures by utilizing a variety of cooling techniques, most frequently by using a cryogenic fluid bath, like liquid helium.

For tissue sectioning, It is a flexible, high-quality device that produces low temperatures. The Greek words “Kryos,” which means cold in English, and “stat,” which means stable, are the roots of the phrase “Cryostat.” The earliest of it was used in the 19th century and are credited to Scottish physicist and chemist Sir James Dewar.

It is also a type of microtome used to cut tissue at extremely low temperatures (usually between 15 and 30 °C) (Figure 55). The chosen tissue section is mounted on a ready-made metal base known as a chuck and covered with a spray of a rapidly freezing substance (such as optimum cutting temperature compound), which is liquid at ambient temperature but soon hardens into a white waxy substance in the cryostat.

Types of Cryostat

Closed-cycle cryostats

Cold helium vapour is pumped through a chamber that makes up closed-cycle Cryostats. The warmer helium exhaust vapour is removed via a mechanical refrigerator outside, where it is cooled and recycled. Compared to other types of cryostats, this uses a lot of electricity.

Continuous-flow Cryostats

Liquid cryogens (usually liquid helium or liquid nitrogen) from a storage Dewar are used to chill continuous-flow Cryostats. The cryogen is continuously refilled by a steady flow from the storage Dewar as it boils inside the Cryostat.

Bath Cryostats

They are built similarly to vacuum flasks that contain liquid helium. The liquid helium bath is placed in thermal contact with a coldplate. Depending on the size and design of the Cryostat, the liquid helium may need to be replaced as it boils away at intervals between a few hours and several months.

Multistage cryostats

Additional cooler stages may be added to the Cryostat to attain temperatures lower than liquid helium at atmospheric pressure. By coupling the coldplate to a 1-K pot, a Hisotope container that can be pushed to low vapor pressure using a vacuum pump, temperatures as low as 1K can be achieved.
